Protein kits for analytical biosurfaces

Create your own analytical surfaces with our tried and tested protein surface assembly kits. Useful for all analytical instruments that use gold substratum e.g. surface plasmon resonance (e.g. Biacore, sensiQ) or quartz crystal microbalance (e.g. Qsense E4, SRS QCM200)

Kits for Antibody Immobilization
We have a range of proteins based on variants of Protein A, G and L that can form monolayers for the immobilization of antibodies of a wide variety of subtypes and species (see Table 1). This gives you choice and flexibility over the range of immunoglobulin binding studies you can do.

The protein kits allow assembly of antibodies in the correct orientation resulting in high specific activity (>60% of immobilised antibody is active for antigen binding compared to 20-25% for amine-coupled antibody). The nature of the monolayer means that non-specific binding is kept to a minimum.

*Orla91 has two protein A domains separated by a spacer sequence that allows binding to both heavy chains, resulting in a higher affinity of binding.

Surfaces made with these proteins may be regenerated by acid wash and reused for binding antibody.

Protein kit components
Each kit contains the following:

  • 3 mL of IgG-binding Orla protein at 10 mM concentration
  • 1 mL of control protein (no IgG-binding domains) at 10 mM
  • 4 mL Filler solution
  • 20 mg of TCEP for protein activation
  • 0.1 mL 50 mM Tris HCl pH7.5
  • 10 mL Buffer ROG-8 protein dilution buffer
  • Protocol

The above kit is enough to prepare 30 IgG-binding and 10 control surfaces on a 2 channel Biacore instrument. If your instrument requires larger injection volumes, call us and we’ll do everything we can to customise the kit to your requirements.
